Book Abstract

流式细胞仪安装有一根或多根激光器，一些低端的流式细胞仪还采用汞光灯作为光源。绝大多数流式细胞仪都配有氩离子气态激光器作为第一根激光，其发射波长为488nm。待测样本中的细胞经液流系统传送，形成单细胞流，依次通过流式细胞仪的流动室，在激光照射区域，细胞上标记的荧光染料受到激光的激发，产生荧光信号。在不同的实验体系中，根据细胞标记的荧光素不同，在不同波长的激光激发下，发射出不同波长的荧光，这些荧光信号可以反映不同的细胞生物学特性。细胞受到激光照射后产生的这些光信号被相应接收器（光电倍增管）接收并放大，转换为与光强度相关的电子信号，然后经计算机储存和处理分析，以图形形式（如直方图、点图、密度图等）直观地显示出细胞的分布情况。流式细胞仪产生的信号主要有散射光信号和荧光信号。这些信号可以反映相应的细胞特征，如反映细胞相对大小的前向散射光（FSC）、反映细胞内部复杂程度的侧向散射光（SSC）以及反映各种细胞功能和抗原表达的荧光信号。将图形和数据直接输入联机专用的计算机，或存人磁盘以备分析。计算机快速而精确地将所测数据进行统计计算，结合多参数分析，从而实现了细胞定量分析。
一些流式细胞仪还配有细胞分选系统，通过细胞分选系统，还可以将具有相同光信号特征，即某些特定特征的细胞群体分选出来。
